New Bacteriophages with Podoviridal Morphotypes Active against Yersinia pestis: Characterization and Application Potential

چکیده
Phages of highly pathogenic bacteria represent an area growing interest for bacterial detection and identification subspecies typing, as well phage therapy environmental decontamination. Eight new phages—YpEc56, YpEc56D, YpEc57, YpEe58, YpEc1, YpEc2, YpEc11, YpYeO9—expressing lytic activity towards Yersinia pestis revealed a virion morphology consistent with the Podoviridae morphotype. These phages lyse all 68 strains from 2 different sets Y. isolates, thus limiting their potential application subtyping but making them rather promising in terms infection control. Two phages—YpYeO9 YpEc11—were selected detailed studies based on source isolation cross other Enterobacteriaceae. The full genome sequencing demonstrated virulent nature phages. Phage YpYeO9 was identified member Teseptimavirus genus YpEc11 Helsettvirus genus, thereby representing species. A challenge assay liquid microcosm YpYeO9/YpEc11 mixture showed elimination EV76 during 4 h at P/B ratio 1000:1. results, combination high lysis stability results culture, low frequency formation resistant mutants, viability under physical–chemical factors indicate practical use antibacterial mean.

Phenotypical characterization of Mongolian Yersinia pestis strains.
Although Mongolia is regarded as one of the possible places of plague radiation, only few data are available from Mongolian Yersinia pestis strains. In this study a total of 100 Mongolian Y. pestis strains isolated from wild mammals and their parasites between the years 1960 and 2007 were analyzed for their phenotype. All strains grew well on selective Cefsulodin-Irgasan-Novobiocin agar and wer...
متن کاملCharacterization of Newly Isolated Lytic Bacteriophages Active against Acinetobacter baumannii
Based on genotyping and host range, two newly isolated lytic bacteriophages, myovirus vB_AbaM_Acibel004 and podovirus vB_AbaP_Acibel007, active against Acinetobacter baumannii clinical strains, were selected from a new phage library for further characterization. The complete genomes of the two phages were analyzed. Both phages are characterized by broad host range and essential features of pote...
متن کاملCharacterization of Yersinia pestis Interactions with Human Neutrophils In vitro
Yersinia pestis is a gram-negative, zoonotic, bacterial pathogen, and the causative agent of plague. The bubonic form of plague occurs subsequent to deposition of bacteria in the skin by the bite of an infected flea. Neutrophils are recruited to the site of infection within the first few hours and interactions between neutrophils and Y. pestis have been demonstrated in vivo. In contrast to macr...
متن کاملYersinia pestis and plague.
Yersinia pestis is the aetiological agent of plague, a disease of humans that has potentially devastating consequences. Evidence indicates that Y. pestis evolved from Yersinia pseudotuberculosis, an enteric pathogen that normally causes a relatively mild disease. Although Y. pestis is considered to be an obligate pathogen, the lifestyle of this organism is surprisingly complex. The bacteria are...
متن کاملIsolation, characterization and therapeutic potential assessment of bacteriophages virulent to Staphylococcus aureus associated with goat mastitis
In the present study, the therapeutic potential of bacteriophages virulent to Staphylococcus aureus associated with goat mastitis were isolated, identified and assessed. Staphylococcus aureus (host or indicator bacterium) was isolated from a goat suffering from clinical mastitis. Based on cultural, morphological, biochemical tests and amplification of S. aureus specific thermonuclease gene inPC...
